However, whether NAD<sup>+<\/sup>&nbsp;depletion occurs in patients with degenerative disorders and whether NAD<sup>+<\/sup>&nbsp;repletion improves their symptoms has remained open. Here, we report systemic NAD<sup>+<\/sup>&nbsp;deficiency in adult-onset mitochondrial myopathy patients. We&nbsp;<strong>administered an increasing dose of NAD<\/strong><strong><sup>+<\/sup><\/strong><strong>-booster niacin, a vitamin B3 form (to 750\u20131,000&nbsp;mg\/day<\/strong>;&nbsp;<a href=\"https:\/\/www.clinicaltrials.gov\/\" target=\"_blank\" rel=\"noreferrer noopener\">clinicaltrials.gov<\/a>&nbsp;<a href=\"https:\/\/clinicaltrials.gov\/show\/NCT03973203\" target=\"_blank\" rel=\"noreferrer noopener\">NCT03973203<\/a>) for patients and their matched controls for 10 or 4&nbsp;months, respectively.&nbsp;<strong>Blood&nbsp;<\/strong><strong>NAD<\/strong><strong><sup>+<\/sup><\/strong><strong>&nbsp;increased in all subjects, up to 8-fold, and muscle NAD<\/strong><strong><sup>+<\/sup><\/strong><strong>&nbsp;of patients reached the level of their controls<\/strong><strong>. Some patients showed anemia tendency, while muscle strength and mitochondrial biogenesis increased in all subjects. In patients, muscle&nbsp;<\/strong><strong>metabolome shifted toward controls and liver fat decreased even 50%<\/strong>.
